Abstract
The present study concerns the experimental evaluation of the influence of chronically administered sexual hormones: Ethinylestradiol, Medroxiprogesterone acetate and estroprogestative oral contraceptives, on Bupivacaine central nervous system toxicity. Bupivacaine is a local anesthetic with an amidic structure. We investigated the convulsant activity of toxic doses of Bupivacaine (50 mg/kg body weight) and that of half the toxic dose (25 mg/kg body weight). We also evaluated the central nervous system effect of Bupivacaine-the usual doses (1 mg/kg body weight) and/or of sexual hormones in daily long term administration, by investigating their influence on the motor activity – the actographic method. The obtained results suggest that pretreatment with estroprogestative oral contraceptives increases the toxicity of Bupivacaine; this pretreatment also stimulates the animals motor activity, as an expression of a central nervous system stimulating effect. Bupivacaine by itself, in the usual dose, inhibits the central nervous system.
